U Myo Win founded SMILE Myanmar (SMILE) in 2007. He has served as the Executive Director of SMILE since its inception. SMILE was founded in response to the increasing intolerance and adoption of unsecular viewpoints by minorities themselves. SMILE began working with religious leaders and school teachers, promoting religious tolerance and civic consciousness, and more participatory learning approaches in educational institutions. 2009 onwards, SMILE expanded its work to work with young people, women and human rights defenders. U Myo Win spoke about the situation of human rights in Burma in 2010, at the Human Rights Sub-committee at the EU, and was the only human rights defender who lived and worked in Burma to speak out. He focused his remarks on security sector reform and for a bottom-up approach to human rights and democracy.
